Description:Alpha-1antitrypsin,Human,mAb3C11,immunoassayantibody
Antibodyclone3C11recognizesboththehealthymonomericform(Mvariant)andthediseaseassociatedpolymericforms(Zvariants)ofhumanalpha-1-antitrypsinwithequalaffinity.Alpha-1-antitrypsinisamemberoftheserineproteaseinhibitor(serpin)superfamilywhichareproteinsknownfortheirABIlitytoinhibitproteases.Itisthemostabundantcirculatingproteaseinhibitorknown.Itmainlytargetsenzymesreleasedbyneutrophils,especiallyneutrophilelastase(NE)butalsoproteinase3(PR3)andCathepsinG(CG).Serpinopathiesareconformationaldiseasescharacterizedbythepolymerizationandintracellularretentionofmembersoftheserpinsuperfamily.Thebestknownisalpha-1antitrypsindeficiency,withthemostcommonseveredeficiencyallelebeingtheZmutation(Glu342Lys).Thissevereautosomaldominantdisordercausestheproteintoundergoaconformationaltransitionandformorderedpolymersthatareretainedwithinhepatocytes.Duetothisaccumulationofpolymersinhepatocytes,bloodalpha-1trypsinlevelswilldecreaseleADIngtochronicuninhibitedtissuebreakdown.Thiscausesthedegradationespeciallyoflungtissuewhichwilleventuallyleadtopulmonaryemphysema.Inaddition,accumulationofpolymersinhepatocytescausesliverdiseasessuchasneonatalhepatitis,cirrhosis,andhepatocellularcarcinoma.
