AntibodycloneCLBVII-1recognizeshumancoagulationfactorVII.FVIIisaserineprotease-coagulatingelementresponsIBLefortheinitiationofanextrinsicpathwayofcoagulation.FVIIisvitaminKdependentandissynthesizedbyhepatocytes.Itisasinglechainglycoproteinofca50kDa,consistingof408aminoacids.Theproteinhasseveralfunctionaldomains,agamma-carboxyglutamicacid-richdomainattheN-terminus,2EGF-likedomainsandtheC-terminalserineproteasedomain.FVIIitselfhasnorealactivity.ActivationofFVIIleadstoFVIIa.Atthesiteofinjury,bloodclottingstartsbyinteractionofFVIIwithtissuefactor(TF).ComplexedwithCa2+,FVIIa/TFactivatesFIXandFX,whichultimatelyleadstotheformationofafibrinclot.
FVIIdeficiencyisararehaemologicaldisorderwithanincidenceof1/500000.Diseaseischaracterizedbymultipleclinicalphenotypes,fromasymptomatictolifethreateningbleedings.FVIIlevelscanbemeasuredbyELISAofturbidimetry,althoughplasmalevelsarelow(0.5μg/ml).Plasmalevelscanbeinfluencedbygenepolymorphisms.
