Description:Calprotectin,Human,mAb27E10,immunoassayantibody
Themonoclonalantibody27E10recognizesanepitopespecificfortheS100A8/A9heterocomplexthatisnotexposedontheindividualsubunitsS100A8(MRP8,calgranulin-A)orS100A9(MRP14,calgranulin-B).Thecalcium-bindingmigrationinhibitoryfactor-relatedproteins,MRP-8(S100A8)andMRP-14(S100A9)belongtotheS100proteinfamily.Theexpressionoftheseproteinsislargelyconfinedtothecytosolofneutrophilsandmonocytes.Thecomplexformationoftheseproteinsisacalcium-dependentprocess.TheS100A8/A9heterocomplex,alsocalledMRP-8/MRP-14complexorcalprotectin,comprises60%ofthecytoplasmicproteinfractionofcirculatingpolymorphonucleargranulocytesandisalsofoundinmonocytes,macrophagesandilealtissueeosinophils.Peripheralbloodmonocytescarrytheantigenextra-andintracellularly,neutrophilsonlyintracellularly.TheS100A8/A9complexhasantibacterial,antifungal,immunomodulatingandantiproliferativeeffects.Besidesthisitisapotentchemotacticfactorforneutrophils.Plasmaconcentrationsareelevatedindiseasesassociatedwithincreasedneutrophilactivity,likeinflammatoryboweldisease.Granulocytesterminatetheirexistenceaftertransmigrationthroughtheintestinalwall.Thereforecalprotectinisalsodetectableinfeces.Elevatedlevelsofcalprotectinhavebeenobservedinbodyfluidssuchasplasma,saliva,gingivalcrevicularfluid,stools,andsynovialfluidduringinfectionandinflammatoryconditions.
Themonoclonalantibody27E10canbeusedforearlydetectionofinflammatorymacrophages,forthecharacterizationoftumoroustissuesandthemonitoringofperipheralbloodcellcultures.Theantibody27E10doesnotreactwithlymphocytesorplatelets.
