MonoclonalantibodyHM23553A3E1recognizeshumanproperdin,alsocalledcomplementfactorP.Thecomplementsystemisthefirstlineofdefenseagainstpathogensandfacilitateseliminationofapoptoticanddamagedcells.Positiveregulatorplasmaproteinproperdiniscriticalforthealternativepathwayofcomplement.Itisasingle-chainglycoprotein(ca53kDa)consistingofsixTSRsequences.Intheblooditexistsasamixtureofpreferablyhead-to-tailtrimers,butalsodimersandtetramers.Theproteinisproducedbyleukocytes,likeactivatedneutrophilsmonocytesandT-lymphocytes,butalsobyeg.stressedendothelialcells.
ProperdincanbothinitiateandpositivelyregulatethealternativepathwayactivitytogetherwithC3andfactorsB,D,IandH.ItbindstoC3bwhereItstABIlizesthelabileC3bBbconvertasewhichisdepositedonimmunecomplexesorforeignsurfaces.TherebyenhancingtheAPbystimulationofamplificationofC3bBb-convertaseformationincompetitionwithcatabolismofC3bbyfactorI,whichusesfactorHasacofactor.ThelocalamplificationprocessleadstothecreationofthealternativepathwayC5convertase,C3bBb3b,andinitiatestheterminalpathwayofcomplementactivation.Thealternativepathwaymayaccountforca80%oftheterminalpathwayactivity.ProperdinhasalsobeenshowntodirectlylimitfactorHactivity.
Recentstudiesshowthatproperdinisalsoapattern-recognitionreceptor(PRR)abletobinddirectlytomicrobialsurfaces,apoptoticandnecroticcells(dangerousnonselfandalteredself).Inappropriateactivationordysregulationofthealternativepathwayisacriticalfactorindevelopmentofseveralautoimmuneconditions.Targetsopsonizedwithproperdinarelabeledforclearancebyscavengercells,evenwithoutcomplement.Thismakesitapotentialtherapeutictargetindiseases.Recentstudieshasshownrenewedinterestintheevaluatingroleofproperdinindiseasepathogenesis,likeAsthma,arthritis,septicshock,AMDandC3glomerulopathy.
Antibody3A3E1canbeusedinELISA,westernblotting(onlynon-reduced)andasantibodyforfunctionalstudiesinhibitingthealternativepathwayofcomplement,likethestandarderythrocyteAP50assay.
