HMGB1(HighMobilityGroupBox-1proteinalsoknownasamphoterin)isahighlyconservedproteinwithmorethan95%aminoacididentitybetweenrodentandhumanHMGB1.Itisaprevalentnon-histonechromatincomponentandanon-sequencespecificDNAbindingprotein.HMGB1consistsoftwohomologousHMGboxesrichinbasicaminoacidsandanacidictailatthecarboxy-terminus.HMGB1isinvolvedintheregulationofchromatinstructureaswellasbeinginvolved,eitheraspositiveornegativefactorswithvariousaspectsofDNAreplication,transcription,repair,andligation.HMGB1,identifiedasamembraneassociatedproteintermed“amphoterin,”mediatesneuriteoutgrowth,tumoroutgrowth,andmetastasis.Itparticipatesinplasminogenactivationandisrecognizedasalatemediatorofendotoxinlethalityinmice.ThemonoclonalantibodyantiHMGB1,clone5H6,canbeusedforWesternblot,FlowcytometryandImmunofluorescence.
