Monoclonalantibody6B11recognizesmousepentraxin3(PTX3).Thefirstlineofdefenseagainstpathogensisrepresentedbytheinnateimmunesystem.Animportantpartofthissystemistherecognitionofpathogenassociatedmolecularpatterns(PAMPs)bypatternrecognitionmolecules(PRMs).Pentraxins(PTXs)areafamilyofsolublecyclicpentamericPRMs.Basedonprimarystructure,theycanbedividedintwogroups,shortandlongPTXs.C-reactiveprotein(CRP)andserumamyloidPcomponent(SAP)areprototypicshortPTXs.OthershavebeenclassifiedaslongPTXs.PTXshaveaconservedca200aminoacidlongc-terminaldomaincontainingthePTXsignature(HxCxS/TWxS,wherexisanyaminoacid).HumanPTX3encodesa381aminoacid,45kDasecretoryglycoproteinwitha162aminoacidN-terminalextensionanda202aminoacidC-terminalpentraxindomain.Thereisalsoahighdegreeofconservationbetweenfrommousetoman.PTX3istheprototypiclongPTXandwasoriginallyidentifiedinendothelialcellsandfibroblasts.OrthologsofPTX3havebeenfoundinavarietyofspecies.ThePTX3geneconsistsofthreeexonsandtwointrons.ThefirsttwoexonscodefortheleaderpeptideandtheN-terminaldomainoftheprotein,respectively,andthethirdexonencodesthepentraxindomain.PTX3expressioncanamongothersbeinducedbyproinflammatorycytokineslikeTNFα,Il1βandTLRagoNISTs.WhereasshortPTXsareproducedintheliver,themainsourceofPTX3expressionaremyeloiddendriticcellsbutexpressionisalsoseeninmonocytes,macrophages,endothelialcells(constitutivelyexpressed),fibroblasts,smoothmusclecells,kidneyepithelialcells,synovialcells,chondrocytes,ADIpocytes,alveolarepithelialcells,granulosacellsandglialcells.PTX3expressioncanbeaffectedbyseveralpathways.PTX3isalsolinkedtothecomplementsystem.ItbindstoC1q,complementfactorHandficolin-2andisthereforeinvolvedintheclassical,alternativeaswellasthelectinpathwayofcomplement.PTX3seemstohaveaimportantregulatingroleofcomplementmediatedimmuneresponses.PTX3actsasanacutephaseproteinandisapotentialMarkerforinfectious,inflammatoryandcardiovascularpathologies.PTX3mayplayaroleinvascularpathology,includingatherosclerosisandrestenosis,andhasbeenconsideredasamarkerofvasculardamage.Inhumans,PTXslevelsarecorrelatedwiththeriskofdevelopingofvascularevents.ItisexpectedthatPTX3haveacardiovascularprotectiveeffect.FurThermore,PTX3upregulationisobservedintheendotheliumfrompatientsaffectedbysystemicsclerosis,adiseasecharacterizedbyinsufficientangiogenesis.Thelatterseemstobemediatedviafibroblastgrowthfactor2(FGF2).Theanti-angiogenicaffectofPTX3,viaFGF2,hasalsopotentialtointerferewithtumorprogression.LikeCRPandSAP,PTX3bindsapoptoticcellsanddebris,andhasbeenassociatedwiththeresponsetotissuedamage.Thisrecognitionofapoptoticcellsissharedbyothercomponentsofinnateimmunity(e.g.collectins).PTX3expressioncanbeaffectedbyseveralpathways.PTX3isalsolinkedtothecomplementsystem.ItbindstoC1q,complementfactorHandficolin-2andisthereforeinvolvedintheclassical,alternativeaswellasthelectinpathwayofcomplement.PTX3seemstohaveaimportantregulatingroleofcomplementmediatedimmuneresponses.PTX3actsasanacutephaseproteinandisapotentialmarkerforinfectious,inflammatoryandcardiovascularpathologies.PTX3mayplayaroleinvascularpathology,includingatherosclerosisandrestenosis,andhasbeenconsideredasamarkerofvasculardamage.Inhumans,PTXslevelsarecorrelatedwiththeriskofdevelopingofvascularevents.ItisexpectedthatPTX3haveacardiovascularprotectiveeffect.Furthermore,PTX3upregulationisobservedintheendotheliumfrompatientsaffectedbysystemicsclerosis,adiseasecharacterizedbyinsufficientangiogenesis.Thelatterseemstobemediatedviafibroblastgrowthfactor2(FGF2).Theanti-angiogenicaffectofPTX3,viaFGF2,hasalsopotentialtointerferewithtumorprogression.LikeCRPandSAP,PTX3bindsapoptoticcellsanddebris,andhasbeenassociatedwiththeresponsetotissuedamage.Thisrecognitionofapoptoticcellsissharedbyothercomponentsofinnateimmunity(e.g.collectins).
