ThemonoclonalantibodyCK-BYK/21E10recognizeshumancreatinekinaseB-type,alsoknownasB-CK.HumanB-CKisaproteinof381aminoacids(~45kDa),expressedinanumberoftissues.B-CKismostabundantinadultbrain,approx.5-foldlowerinthestomach,10-foldlowerintheheartandbarelydetectableinliver.Inbrain,whereasmostB-CKhasbeenshowntobecytosolic,severalofthereactionsrequiringB-CKaremembrane-associated.B-CKbelongstothecreatinekinase(CK)isoenzymesthatcatalysethesynthesisofphosphocreatine(PCr)anditssubsequentuseintheregenerationofATPincelltypeswheretheconsumptionofATPisrapidand/orsudden.InthebrainthedifferentCKisoformsconstituteanenergyshuttlewhereinATPproducedinthemitochondriaisusedbyamitochondrialCK[e.g.ubiquitousmitochondrialcreatinekinase(uMi-CK)]togeneratePCr,whichisthentransportedandusedbyacytoplasmicCK[e.g.braincreatinekinase(B-CK)]toregenerateATPatdiscretecellularsitesofhighATPturnover.B-CKappearstohavearoleinregeneratingATPneededforthetransportofionsandneurotransmitterssinceCKBhasbeenlocalizedtobrainsynapticplasmamembranes,possIBLycoupledtoNa+/K+-ATPaseandacetylcholinereceptor-richmembranes.
