ThemonoclonalantibodyHM.1recognizeshumanImmunoglobulin-likeCellAdhesionMolecule-1(ICAM-1).ThehumanICAM-1proteinconsistof505aminoacidsfunctioningasatypeItransmembraneproteinbelongingtotheimmunoglobulinsuperfamily,C2subset.ItcontainsfiveIg-likedomainsinitsextracellularpart,atransmembranedomainandachargedcytoplasmictail.ICAM-1isdifferentiallyglycosylateddependingoncelltype,givingrisetoan80-114kDaprotein.DeglyosylatedICAM-1is~60kDa.ICAM-1isexpressedbyawidevarietyofcellsincludingendothelialandepithelialcells,macrophages,fibroblast-likecellsanddendriticcells.ICAM-1expressioncanbeinducedbyinflammatorycytokines(interleukin-1(IL-1),Interferongamma(IFNα),tumornecrosisfactoralpha(TNFα)),phorbolestersorlipopolysaccharide.ExpressionregulationiscellspecificanddependsontheavailABIlityofcytokine/hormonereceptors,signaltransductionpathways,transcriptionfactors,andposttranslationalmodification.ICAM-1isanessentialcomponentinmanyimmune-relatedprocesses.ICAM-1linkswithreceptorsoftheintegrinfamily,therebymediatingcell-cellinteractionsandallowingforsignaltransduction.ICAM-1interactsspecificallywithitsreceptorstoinduceareversIBLeadhesioninteraction.ForprocesseslikeTcellactivationandleucocyterecruitment,normalimmunefunctionreliesonICAM-1.LigandsforICAM-1aretheIntegrinsLFA-1andMAC-1onleukocytes,CD43(sialophorin),thesolublemoleculefibrinogen,thematrixfactorhyaluronan,rhinoviruses,andPlasmodiumfalciparummalaria-infectederythrocytes.DerangementofICAM-1expressioncontributestotheclinicalmanifestationsofavarietyofdiseases,predominantlybyinterferingwithnormalimmunefunctions.Amongthesearemalignancies,manyinflammatoryconditions(e.g.asthma,autoimmunedisorders),atherosclerosis,ischemia,certainneurologicaldisorders(e.g.multiplesclerosis)andallogeneicorgantransplantation.ICAM-1isshedbythecellanddetectedinplasma,CSFand/orBALassolubleICAM-1(sICAM-1).sICAM-1canbeveryusefulasbioMarkerfordiseaseactivity.
