Description:PAI-1,Rat,mAbMA-124K1
Plasminogenactivatorinhibitortype-1(PAI-1),amemberoftheserineproteaseinhibitor(serpin)superfamilyisanimportantproteinintheregulationoffibrinolysis.PAI-1isuniqueamongtheserpinsbecauseofitsfunctionalandconformationalflexibility.PAI-1isthemostimportantphysiologicalinhibitorofbothtissue-typeplasminogenactivator(t-PA)andurokinase-typeplasminogenactivator(u-PA).IncreasedPAI-1levelsareassociatedwiththromboticeventsandisanestablishedriskfactorforcardiovasculardiseases.TheactiveconformationPAI-1inhibitsitstargetproteinasesbytheformationofastable,inactivecomplex.AlthoughPAI-1issynthesizedasanactivemolecule,itconvertsspontaneouslytoaninactive,latentformthatcanbepartiallyreactivatedbydenaturingagents.Inaddition,athirdconformationreactingasanon-inhibitorysubstratetowardsvarioustargetproteinaseshasbeenidentified.TheepitopeofmonoclonalantibodyMA-124K1isaconformationalepitopeclosetothereactive-siteloop.Glu220andGlu212constitutethemainmoleculardeterminantsoftheepitopeforMA-124K1.ThemonoclonalantibodyMA-124K1inhibitsPAI-1activityandsimultaneouslyincreasesthebindingofinactivePAI-1tovitronectin.Theeffectoftheantibodyinvolvesanallostericmodulationofthevitronectinbindingsite.TheantibodycrossreactswithmousePAI-1.
