Themonoclonalantibody5C11recognizeshumanADAMTS-13,ADisintegrinAndMetalloproteasewithThromboSpondintype1domain13.ADAMTS-13isproducedbyhepaticstellatecellsandinsmalleramountsbyhumanendothelialcells,andispresentinplasmaataconcentrationofapproximately1ug/ml.ADAMTS-13isazinc-containingmetalloproteasebelongingtotheADAMTSfamilycharacterizedbyaproteasedomain,anadjacentdisintegrin-likedomain,oneormorethrombospondintype1repeats,acystein-richdomainandatypicalspacerregion.ADAMTS-13iscomposedofaseriesofdomains(aminotocarboxyterminal):metalloprotease,disintegrin-like,centralthrombospondin-1(TSP-1),cysteine-rich,spacer,sevenadditionalTSP-1domainsandtwouniqueCUBdomains.ADAMTS-13hasnohydrophobictransmembranedomain,andhenceitisnotanchoredinthecellmembrane.Theapparentmolecularweightis170or190kDaonnon-reducingorreducingSDS-PAGE,respectively.ADAMTS-13hasanimportantfunctioninhaemostasis,whereitcatalyzesthecleavageofthepeptidebondbetweentyrosine-1605andmethionine-1606intheA2domainofvonWillebrandFactor(VWF),resultingin2electrophoreticreducedfragmentsof176and140kDa,respectively.Thisprocessrenderslargemultimerslessadhesiveandhencelessreactiveinthesettingofthrombusformation.ADAMTS-13isthereforesaidtobeanaturalanti-thromboticagent.SevereADAMTS-13deficiencyisassociatedwithsystemicmicrovascularthrombosisinfamilialoracquiredthromboticthrombocytopenicpurpura(TTP).Theaccumulationofnon-cleavedlargeVWFmultimerscausesspontaneoussystemicplateletaggregationblockingoxygensupplytovitalorgans.Thislife-threateningdisordercanleadtoischemicdiseasewith(multiple)organfailure.Themonoclonalantibody5C11recognizesthecentraltoC-terminalTSP-1repeats2to5ofADAMTS-13(aminoacid686-894).
