ThemonoclonalantibodyDCN47.5reactswiththeC-typelectin,DC-SIGN(CD209),exclusivelyexpressedonhumandendriticcells(DC).DCarespecializedantigenpresentingcellsandbridgetheinnateandtheadaptiveimmunesystem.Theyprovidehighlevelsofcostimulationnecessaryforactivationofbothnaïveandantigen-experiencedT-cells.ImmatureDCarecapabletomigratetoinflammatorysites,captureantigenandprocessitinternallytoformMHC-peptidecomplexes.Followingantigenuptake,DCundergomaturationandmigratetolymphoidorganswheretheycanpresentMHC-peptidecomplexestorestingT-cellsanddriveT-cellproliferation.DuringdifferentiationandmaturationofDC,severalphenotypicsurfaceMarkersareexpressed:CD1a,CD4,CD11,CD40,CD86,andHLA-DR.ImmatureDCpredominantlyexpressCCR5whichenablesDCtomigratetoinflammatorysites,whereasmatureDCexpresshighlevelsofCXCR4,areceptorthatfacilitatesmigrationtolymphoidorgans.
DCalsoexpressDC-specific,ICAM-3grabbing,nonintegrin(DC-SIGN),a44kDaC-typelectinthatbindstotheHIV-1envelopeglycoproteingp120,ICAM-3onT-cellsandICAM-2onendothelialcells.HIVvirionsareabletoinfectcellsexpressingCD4andthechemokineco-receptorsCCR5orCXCR4andcanattachtoDC-SIGNtoextendvirionLifespan.Therefore,DCarecandidatesforHIV-1infection.DC-SIGN-ICAM-3bindingisintegrin-independentbutcalcium-dependentandantibodiesreactiveagainstDC-SIGNcanbeusedtostudyDC-SIGN-ICAM3binding.ThemonoclonalantibodyDCN47.5specificallyreactswiththeC-typelectinDC-SIGN(CD209)expressedonhumandendriticcellsandinhibitsbindingofDC-SIGNtoICAM-2onendothelialcells.
