Themonoclonalantibody4recognizesthecleavedhumanC3fragmentC3c.Thecomplementsystemisanimportantfactorininnateimmunity.Thethirdcomplementcomponent,C3,iscentraltotheclassical,alternativeandlectinpathwaysofcomplementactivation.Activationproductsofthecomplementcascadecontainneo-epitopesthatarenotpresentintheindividualnativecomponents.ThesynthesisofC3istissue-specificandismodulatedinresponsetoavarietyofstimulatoryagents.C3isthemostabundantproteinofthecomplementsystemwithserumproteinlevelsofabout1.3mg/ml.AninheriteddeficiencyofC3predisposesthepersontofrequentbacterialinfections.C3fragmentsaredepositedintissuesatsitesofantibody-mediatedimmunopathology.Inulcerativecolitisandidiopathicchronicinflammatoryboweldisease,thedepositionofC3inthediseasedmucosahasbeenreported.ProteolysisbyC3-convertasesresultsinthecleavageofC3intoC3aandC3b.C3bbecomesattachedtoimmunecomplexesandisfurthercleavedintoiC3bandC3f.iC3bisfurtherprocessedintoC3candC3dg.Themonoclonalantibody4recognizesaconformationalepitopeinC3c,C3bandiC3b.
