Themonoclonalantibody697reactswithhumanneutrophillipocalin(HNL),alsoknownashumannGAL(25kDa).Lipocalinsareawidespreadfamilyofsmall,robustproteinsthattypicallytransportorstoreBIOLOGicalcompoundswhichareeitheroflowsolubilityorarechemicallysensitive,includingvitamins,steroids,odorantsandmetabolicproducts.Themembersofthislipocalinsshareahighsimilarityintheirtertiarystructuresinspiteofalowdegreeinamino-acidsequenceidentity:threehighlyconservedsequencemotifsformafunnel-likebetabarrelwhichenclosesahydrophobicpocketfortheinternalligandbinding.Thehumanneutrophillipocalin(HNL)isproducedbyneutrophilleukocytesandstoredinintracellularspecificgranules.NeutrophillipocaliniscovalentlylinkedtogelatinaseB(MMP-9)andproMMPisstoredinthreeforms:themonomericform(92kDa),thehomodimer(220kDa)andaheterodimer(125kDa).Duringinflammationandneoplastictransformationthehumanneutrophillipocalin(HNL)isproducedinneutrophilsandinepithelialcells.FurThermoreneutrophillipocalinisasuperiormeanstodistinguishacutebacterialandviralinfectionsaswellasitenablesthedeterminationoftheneutrophilactivationlevel.
