Monoclonalantibody1837-46reactswithendostatin,a20kDaC-terminalfragmentofcollagenXVIII.Endostatinisabletoevokenon-uniformresponseforproliferation,cellmountandmigrationofentothelialcells,withdifferentendostatinbindingcharacteristic,leadstotheassumptionthatendostatineffectisstronglydependentfromendothelialcelltype.FurThermoreendostatininhibitsangiogenesisandtumorgrowthinvivobyinducingapoptosisinendothelialcells.Thelocaldeliveryofendostatinseemstospecificallyaffecttumor-associatedmicrovesselsbyreductionofthevesseldensity,diameterandfunctionality.Tumorcellmigrationandinvasionwasgreatlyreducedintheendostatintreatedanimals.Endostatinisnon-toxicanddoesnotinduceacquireddrugresistanceandhasthereforebecomeapotentnewtherapystrategyinsolidneoplasias.Thistherapyappeartohavehighpotentialnotonlyforthetreatmentofgliomas,themostcommonbraintumours,butalsoofothertumours.TheABIlityofendostatintoinhibitneoangiogenesisismediated,atleastinpart,byZn2+bindingandelastaseprocessing.Widespreadendostatinexpressionwasfoundinelasticfibersinvesselwallsandinsomeotherbasementmembranezones.EndostatinisreleasedbyneuronstoaccumulateinamyloidplaquesinAlzheimer"sdisease.
