ThemonoclonalantibodyB2C10reactswithgalectin-3,a30kDaprotein.Galectin-3isamemberofthegalectinfamily.Theproteiniscomposedofthreedomains:asmallamino-terminaldomain,acarboxyl-terminalcarbohydraterecognitiondomain(CRD)andamino-terminaldomaincontainingrepeatingelements.Galectin-3isnormallydistributedinepitheliaofmanyorgansandvariousinflammatorycells,includingmacrophages,aswellasdendriticcellsandKupffercells.Theexpressionofthislectinisup-regulatedduringinflammation,cellproliferation,celldifferentiationandthroughtrans-activationbyviralproteins.Theexpressionisalsoaffectedbyneoplastictransformation:up-regulatedincertaintypesoflymphomasandthyroidcarcinoma,whiledown-regulatedinothertypesofmalignancies,suchascolon,breast,ovariananduterinecarcinomas.
Galectin-3hasbeenshowntofunctionthroughbothintracellularandextracellularactions.Relatedtoitsintracellularfunctions,galectin-3hasbeenidentifiedasacomponentofheterogeneousnuclearribonuclearprotein(hnRNP),afactorinpre-mRNAsplicing,andhasbeenfoundtocontrolcellcycleandpreventTcellapoptosis.Ontheotherhand,thisproteinhasalsobeendemonstratedtofunctionasextracellularmoleculeinactivatingvarioustypesofcells,includingmonocytes/macrophages,mastcells,neutrophilsandlymphocytes.Galectin-3hasbeenshowntomediatecell-cellandcell-extracellularmatrixinteractions.ThemonoclonalantibodyB2C10inhibitsthebindingof125I-labeledgalectin-3toIgEcoatedonmicrotiterplates,thegalecin-3’shemagglutinationactivityandgalectin-3-inducedsuperoxideproductionbyhumanneutrophils.ThisinhibitoryactivityofB2C10isprobablytheresultofitsdisruptionoftheself-associationprocess.
TheepitopeofthemonoclonalantibodyB2C10isfoundwithinthefirst45aminoacidsofgalectin-3.TheantibodyB2C10doesnotreactwithGalecin-3Candiscrossreactivewithmousegalectin-3.
