Monoclonalantibody546-2reactswithendothelial-monocyte-activatingpolypeptideII(EMAPII),a20kDapolypeptide.EMAPIIisanantiangiogenicandproinflammatorycytokine(expressedbymicroglia/macrophages).EMAPIIappearstobeaprofoundmodulatorofinflammatoryreactionsoftheinnateimmunesystemandmightparticipateinavarietyofpathogenicimmune-processesinthemammalianbrain.EMAPIIisapotentialmediatorofinflammatoryresponsesinautoimmunityandaconsideredtargettostagereactionsofmonocyticlineagecellsininflammatoryreactions.InnormaltissuesEMAPIIisexpressedinendocrineorgans,incellsofneuroendocrineorigin,butalsointissueswithhighcellturnover.UnderpathophysiologicalconditionsEMAPIIhasseveralfunctions.Ittriggerstherecruitmentofmacrophagesbyelevatingcytosolic-freeCa2+concentrations,itstimulatesleukocytechemotaxis,itinducestheexpressionofTNF-alphaandtissuefactor(TF)bymonocytesandmyeloperoxidase(MPO)bypolymorphonuclearcells(PNMs).FurThermoreitstimulatesthereleaseofvonWillebrand-factor,P-andE-selectinbyendothelialcellsandEMAPIIinducesendothelialprogrammedcelldeathinvitro.Themonoclonalantibody546-2iscrossreactivewithratendothelial-monocyte-activatingpolypeptideII(ratEMAPII).
