Monoclonalantibody1022-5reactswithAllograft-inflammatoryfactor-1(AIF-1)polypeptide,alsoknownasIonizedcalcium-bindingadaptermolecule1,Iba-1.AIF-1isa17kDaIFN-gammainducIBLeCa2+-bindingEF-handprotein,associatedwithchronicinflammatoryprocesses.AIF-1isconstitutivelyexpressedinlymphatictissueandisup-regulatedinmonocytes/macrophagesundervariouspathologicalconditionssuchaschronicrejectionofallograftsandinautoimmunelesions.Inthebrain,asubsetofmicroglialcellsconstitutivelyexpressAIF-1.IncreasednumbersofAIF-1immunoreactivemacrophage/microglialcellswereobservedinfocalbraininfarctions,traumaticbraininjury,gliomas,uveitisandininflammatorylesionsofaratmodelofautoimmunedisease,autoimmuneencephalomyelitisandneuritis.SeveralCa2+-bindingpeptideshavebeenassociatedwithmonocyteactivation,inparticularmembersoftheS-100supergene-family.AIF-1isaCa2+-bindingpeptidewhichisnotamemberoftheS-100geneclusterbutislocatedwithintheHLAclassIIIgenomicregion.Themonoclonalantibody1022-5iscrossreactivewithratAllograft-inflammatoryfactor-1(ratAIF-1).Humanandrataminoacidssequencesare98%homologous.
