Themonoclonalantibody24recognizesanepitopeof174kDpresentonleukocyteintegrins,CD11/CD18(LFA-1)orbeta2-typeintegrins.Theleukocyteintegrinsareafamilyofheterodimericreceptorsthatmediatedivalentcation-dependentcellularadhesionreactions.Tcellsuseintegrinsinessentiallyalloftheirfunctions.Integrinsbecomeactivefollowingsignallingthroughothermembranereceptors,whichcausebothaffinityalterationandanincreaseinintegrinclustering.Themonoclonalantibody24recognizesastructuralfeatureandisstrictlydependentuponthepresenceofMg2+.Theepitopeislocatedwithin,orincloseproximityto,thethreeconservedcationbindingdomainsandthereforeameasureofMg2+boundtotheleukocyteintegrinsandthusreflectsfunctionallyactivemolecules.Theepitopecanbeinducedonpolymorphonuclearleukocytesandonmonocytes.Glu173andGlu175ofthebeta(2)Idomainareidentifiedascriticalforantibody24binding.Themonoclonalantibody24inhibitsmonocyte-dependent,antigenspecificTcellproliferationandIL-2-activatednaturalkillercellassays.Theantibodydoesnotinterferewithmitogen-stimulatedTcellproliferation.FurThermorethemonoclonalantibody24prevents“deadhesion”ofreceptor/ligandpairs,possIBLelockingleukocyteintegrinsinan“active”conformation.
