Plasminogenactivatorinhibitortype-1(PAI-1),amemberoftheserineproteaseinhibitor(serpin)superfamilyisanimportantproteinintheregulationoffibrinolysis.PAI-1isuniqueamongtheserpinsbecauseofitsfunctionalandconformationalflexibility.PAI-1isthemostimportantphysiologicalinhibitorofbothtissue-typeplasminogenactivator(t-PA)andurokinase-typeplasminogenactivator(u-PA).IncreasedPAI-1levelsareassociatedwiththromboticeventsandisanestablishedriskfactorforcardiovasculardiseases.TheactiveconformationPAI-1inhibitsitstargetproteinasesbytheformationofastable,inactivecomplex.AlthoughPAI-1issynthesizedasanactivemolecule,itconvertsspontaneouslytoaninactive,latentformthatcanbepartiallyreactivatedbydenaturingagents.Inaddition,athirdconformationreactingasanon-inhibitorysubstratetowardsvarioustargetproteinaseshasbeenidentified.TheresiduesGlu242,Lys243andGlu244,locatedintheloopconnectings3BwithhG(designatedloop2)andtheresiduesGlu350,Asp355andArg356,locatedatthedistalhingeofthereactivesiteloop,formthemajorsiteofinteractionforthemonoclonalantibodyMA-56A7C10.Thisisclearlyaconformationalepitopecomprisingresiduesclusteredtogetherinthethreedimensionalstructure.MA-56A7C10exhibitsdifferentialreactivitytowardthedifferentconformationsofhumanPAI-1ThemonoclonalantibodyMA-56A7C10generatesanon-reactiveformofPAI-1,whetherthisisthelatentformorthatthereactivesiteisblockedbytheantibodyisunknown.TheantibodycrossreactswithporcinePAI-1.
