Plasminogenactivatorinhibitortype-1(PAI-1),amemberoftheserineproteaseinhibitor(serpin)superfamilyisanimportantproteinintheregulationoffibrinolysis.PAI-1isuniqueamongtheserpinsbecauseofitsfunctionalandconformationalflexibility.PAI-1isthemostimportantphysiologicalinhibitorofbothtissue-typeplasminogenactivator(t-PA)andurokinase-typeplasminogenactivator(u-PA).IncreasedPAI-1levelsareassociatedwiththromboticeventsandisanestablishedriskfactorforcardiovasculardiseases.TheactiveconformationPAI-1inhibitsitstargetproteinasesbytheformationofastable,inactivecomplex.AlthoughPAI-1issynthesizedasanactivemolecule,itconvertsspontaneouslytoaninactive,latentformthatcanbepartiallyreactivatedbydenaturingagents.Inaddition,athirdconformationreactingasanon-inhibitorysubstratetowardsvarioustargetproteinaseshasbeenidentified.TheepitopeofmonoclonalantibodyMA-55F4C12iswithinthestretchof29aminoacidsinPAI-1(regionGlu128-Ala131).Glu128,Val129,Glu130,Arg131andLys154arethemajordeterminants.AllfiveresiduesactcooperativelyinthebindingtoMA-55F4C12andconstitutetheepitope.Theepitopeoftheantibodydoesnotcoverthecompletealpha-helixFandturnconnectingalpha-helixFandbeta-strands3A,butisrestrictedtothehingeregionbetweenalpha-helixFandthemainpartofthePAI-1molecule.ThemonoclonalantibodyMA-55F4C12isa"switching"antibody,capableofinducinganon-inhibitorysubstrateformofPAI-1.TheantibodycrossreactswithratandmousePAI-1.
