Themonoclonalantibody265-1K1reactswithhumanlactoferrin(LF),an80kDaglycoprotein.Lactoferrinwasfirstisolatedfromhumanmilkandplaysanimportantpartintheimmunesystemandhelpstofightinfections.Lactoferrinpromotesthehealthofthegastro-intestinalsystembyimprovingtheintestinalmicrobialbalance.Inaddition,LFcanbefoundinepitheliaandmostbodyfluidsandsecretions.Lactoferrinissecretedinplasmabyneutrophils.Itsplasmaconcentrationalsorepresentsapositiverelationtothetotalpoolofneutrophilsandtherateofneutrophilturnover.Ininflammationlactoferrinisreleasedfromsecondarygranulesofneutrophilicleukocytesintotheextracellularmedium.Thereforetheextracellularlactoferrinconcentrationcanbeusedasanindexforneutrophilactivation.Lactoferrinstronglybindstoironandthisironbindingpropertyisconsideredtobeanimportantantimicrobial.HumanlactoferrinbindstobacterialproductsthroughitshighlypositivelychargedN-terminus,itkillsvariousbacteria,mostprobablybyinducingintracellularchangesinthesebacteriawithoutaffectingthemembranepermeABIlity.CleavagebypepsinoflactoferrinleadstothereleaseoflactoferricinH.This47aminoacidpeptidehasmoreantimicrobialactivitythanitsprecursoranditcaninhibittheclassicalbutnotthealternativecomplementpathway.Lactoferrinalsoplaysaroleinsignaltransduction,immunomodulationandhasantiadhesive,anticancer,antiviralactivity.
