MonoclonalantibodyPR3G-2reactswithhumanproteinase3(PR3),a30kDaprotein.PR3isamajorantigenrecognizedbyautoantibodiesdirectedagainstcytoplasmicproteinsofneutrophilicgranulocytesandmonocytes(calledanti-neutrophilcytoplasmicautoantibodies(ANCA)).ANCAareabletoactivateprimedneutrophilstoproduceoxygenrADIcalsandreleaselyticenzymes,includingPR3.Proteinase3(PR3)wasidentifiedasthetargetantigenofANCAinWegener"sgranulomatosis(WG).ANCAdirectedagainstPR3(PR3-ANCA)caninterferewiththebindingofPR3toitsphysiologicalinhibitoralpha1-antitrypsin(alpha1-AT)andwiththeproteolyticactivityofPR3.Atthesiteofinflammation,PR3cancleavethePR3-ANCAcomplexbetweentheseinhibitingANCAandPR3itself,leavingactivePR3.AutoantibodiestoPR3arepotentactivatorsofthe5-lipoxygenasepathwayinprimedhumanneutrophils.Extracellularfreearachidonicacid,aspresentataninflammatoryfocus,synergizeswithsuchautoantibodiestoevokefull-blownlipidmediatorgeneration,granulesecretionandrespiratoryburst.Proteinase3(PR3)isaneutralserineproteinase,whichislocalizedintheazurophilicgranulesofneutrophilsandingranulesofmonocytesandcanbedetectedinthemembraneofsecretoryvesicles.PR3degradesanumberofextracellularmatrixproteinssuchaselastinandinactivateshumanC1inhibitor.Membrane-associatedPR3isalsoabletoactivatecaspase-3withouttriggeringapoptosisofneutrophils,whichispossIBLyaneutrophilsurvivalmechanism.Inaddition,PR3isinvolvedinmyeloiddifferentiationandis,therefore,alsocalledmyeloblastin.ThemonoclonalantibodyPR3G-2wasproducedbyimmunizationofmicewithacrudegranuleextract.
