Monoclonalantibody1B2-8-2reactswithPunctin-1(ADAMTSL-1).Punctin-1isasecretedmoleculeresemblingmembersofthefamilyofproteasesdesignatedADAMTS(adisintegrin-likeandmetalloproteasedomainwiththrombospondintypeImotif).ADAMTSproteasesaresecretedenzymesthatconsistofametalloproteasedomain(whichprovidesthecatalyticactivity)andancillarydomains(whichprovidesubstrate,cellorextracellularmatrixactivity).Punctin-1lacksthepro-metalloproteaseandthedisintegrin-likedomaintypicalofthisfamilybutcontainsotherADAMTSdomainsinpreciseorderincluding4thrombospondin-Irepeats.Punctin-1isanextracellularmatrixproteinintissuesanddepositedintothecellsubstratumandlocalizedtowardstheperipheryoftransfectedcells.Punctin-1andPunctin-2(ADAMTSL-3)arethefirstmembersofanovelfamilyofADAMTS-likeproteinsthatappeartohaveimportantfunctionsintheextracellularmatrix.Monoclonalantibody1B2-8-2recognizesthefirstthrombospondinrepeatneartheN-terminalandreactswithmolecularspeciesof55kDaoradoubletof55kDaand60kDa(glycosylatedform).NotethatproteolysisofPunctin-1hasbeendescribedandadditionalsmallerbandsmayalsobeobtained.
