Themonoclonalantibody44C143reactswithhumanToll-likereceptor8(120kDa).TLRsbelongtoafamilyofproteinsthatspecificallyrecognizesandsensesmicrobialproducts.Theyarehighlyconservedthroughoutevolutionandhavebeenimplicatedintheinnatedefensetomanypathogens.LikeitscounterpartsinDrosophilatollisrequiredfortheanti-fungalresponse,whiletherelated18-wheelerisinvolvedinantibacterialdefenses.Inmammals,TLRareidentifiedastypeItransmembranesignalingreceptorswithpatternrecognitioncapABIlities.ThevariousTLRsexhibitdifferentpatternsofexpression.TheTLR8genelislocatedincloseproximitytoanotherfamilymember,TLR7,onhumanchromosomeX.TheTLR8geneconsistsofthreeexons,twoofwhichhavecodingfunction.TLR3,7,8and9belongtoathirdclassofTLRswhicharelocalizedintracellularwheretheydetectnucleicacidsderivedfromvirusesandbacteria.TLR8recognizesimidazoquinolinesandssRNAandinteractswithitsligandsinendosomal/lysosomalcompartments.SmallinterferingRNA(siRNA)canactivateTLR8onlywhenmRNAiscomplexedwithlipofectinandwhenthesiRNAsareencapsulatedintoliposomes.NKcellsareactivatedthroughTLR7/8andreportacentralroleformonocyte-derivedIL-12inthedifferentialIFN-producedbyNKcellsinresponsetodifferentTLRagoNISTs.
