The monoclonal antibody 19D759.2 recognizes an intracellular epitope in the cytoplasmic domain of
human TLR5. TLRs belong to a family of proteins that specifically recognizes and senses microbial
products. They are highly conserved throughout evolution and have been implicated in the innate
defense to many pathogens. In Drosophila toll is required for the anti-fungal response, while the related
18-wheeler is involved in antibacterial defenses. In mammals, TLR identified as type I transmembrane
signaling receptors with pattern recognition capabilities. The various TLRs exhibit different patterns of
expression.
TLR5 is upregulated in response to exposure to bacteria or the TLR5 agonist, flagellin. Gram-negative
bacteria stimulate monocyte/macrophage cells in a TLR5 specific, CD14-independent manner.
Signaling through the TLR5 receptor appears to be the way by which the innate immune system
recognizes flagellated bacterial pathogens. It acts via MYD88 and TRAF6, leading to NF-kappa-B
activation, cytokine secretion and the inflammatory response.
Genetic variation in TLR5 is associated with resistance to systemic lupus erythematosus type 1
(SLEB1).
