Themonoclonalantibody25F9recognisesaproteinof86kDonthecellsurfaceandwithinthecytoplasmofmaturemacrophages.Theantibodyisassociatedwithfullydifferentiatedtissuemacrophagesbothinnormalandindiseasedtissues,independentlyofthepresenceorabsenceofinflammation.Theantigenisabsentonfreshlyisolatedmonocytesandotherbloodcells.After6to7daysculturehumanmonocytesbecomepositive.Somemelanomaandcarcinomacelllinesarealsopositive.FurThermorethemonoclonalantibody25F9crossreactswithasubpopulationofmacrophagesofrhesusmonkey,pigalveolarmacrophagesandKupffercells.Themonoclonalantibody25F9isveryusefulformacrophagephenotyping.
