ThemonoclonalantibodyB4/7recognizeshumanandcanineguaninenucleotideexchangefactorH1(GEF-H1).GEFH1isan~110kDaproteinbelongingtotheDblfamilyofproto-oncogenes.GEF-H1canactivatethesmallGTPaseRhoA,butnotRac1orCdc42.RhofamilyGTPasesarecentralregulatorsofepithelialtightjunctionsandtheCytoskeleton.GEF-H1canassociatewithdifferentcytoskeletalstructures,namelymicrotubulesandtheactincytoskeleton.Ithasalsobeenproposedtomediatecrosstalkbetweenthetwotypesoffilaments.
LocalizationofGEF-H1differsbetweencelltypes.InMRC-5fibroblastcells,GEF-H1localizestostressfibers.Inepithelialcells,GEF-H1isassociatedwithapicaltightjunctionsandinvolvedinregulatingparacellularpermeABIlityofsmallhydrophilictracers.FurThermore,itssubcellularlocalizationchangesinmitoticcells,whereendogenousGEF-H1isconcentratedatmitoticspindles.GEF-H1iscapableofbindingtotheF-actinbindingjunctionaladaptorcingulin.BindingtocingulininhibitsGEF-H1andresultsinthedownregulationofRhoAandinhibitionofG1/Sphasetransition.Inlowconfluentculturedcells,thelocalizationofGEF-H1ispredominantlycytoplasmic.Withincreasingdensityofthecells,freeGEF-H1issequesteredatthetightjunctionsbycingulin.
GEF-H1ispartofthesignalingpathwayconnectingepithelialpolaritywiththecellcycle,andassuchinvolvedinoncogenesis.
