MonoclonalantibodyPS7isspecificforhumanProteinS(PS),avitaminK-dependentplasmaglycoproteinwithamolecularweightofapproximately70kDa.Inhumans,PSismainlysynthesizedbyhepatocytes.MaturePShasamodularstructureconsistingofaGla-domaincontainingashortaromaticstack,aregionsensitivetocleavagebythrombinandfactorXa,4epidermalgrowthfactor(EGF)-likedomains,andasexhormone-bindingglobulin-likeregion.TheBIOLOGicfunctionofPSthoughttobeofprimaryimportanceisitsABIlitytoenhanceActivatedProteinC-dependentproteolyticinactivationoffactorsVaandVIIIa,whicharerespectivelythecofactorsinthepro-thrombinaseandtenasecomplexesofthecoagulationcascade.Inthisway,PSplaysanimportantroleinregulatingthrombingeneration,andthereforecontrollingprocoagulantactivity.CrucialtotheAPC-dependentfunctionsofPSisthehigh-affinityinteractionbetweenPSandnegativelychargedphospholipidsurfaces,conferredbycalcium-inducedfoldingoftheamino-terminalGla-domain.MonoclonalantibodyPS7allowsspecificdetectionofcellscarryingPSbyflowcytometry.
