ThemonoclonalantibodyE9reactswithEndoglin,a190kDahomodimerictransmembraneglycoproteincomposedofdisulfide-linkedsubunits.TheexternaldomainbindsTGF-beta1and-beta3isoformswithhighaffinity.Twodifferentisoforms(LandS)ofCD105withcapacitytobindTGF-betahavebeencharacterized,whichdifferintheaminoacidcompositionoftheircytoplasmictails.Mutationsinthegeneencodingendoglinhavebeenlinkedtothehumandiseasehereditaryhemorrhagictelangiectasiatype1(HHT1),avasculardisordercharacterizedbylocalizedvasculardysplasiaandatendencytowardsarteriovenousmalformations.MiceexpressingasingleCD105alleledevelopexternalsignsofdiseasesimilartohumanHHT1,supportingthehaploinsufficiencymodelforHHT1.Micelackingendoglindiefromdefectiveangiogenesischaracterizedbyfailureofvascularsmoothmuscleinvestmentofembryonicbloodvessels,suggestingadefectinvascularsmoothmusclecelldevelopment.Microvesseldensity(MVD)hasbeenreportedtobeanindependentprognosticindicatorofoutcomeinavarietyofhumanmalignancies,withincreasedMVDcorrelatingwithshorteroverallandrelapse-freesurvivalrates.TheMVDcountsusinganti-CD105antibodysignificantlycorrelatedwithoverallanddisease-freesurvival.Anti-CD105monoclonalantibodyE9andanti-CD34monoclonalantibodyhavebeensuccessfullyusedtoquantifyMVDinhumanbreastcarcinoma.ThemonoclonalantibodyE9,directedagainstCD105,hasalsobeenusedasaprognosticMarkerforprimarycentralnervoussystemlymphomas.
