ThemonoclonalantibodyF4B2-B11reactsspecificallywiththeLIM-onlyproteinFHL2.Fiveproteinssharethesamestructuralorganizationandahighdegreeofsequencehomologyinthisgroup:fourandahalfLIMdomainprotein(FHL)1,FHL2,FHL3,FHL4,andactivatorofcAMP-responsiveelement(CRE)modulator(CREM)intestis(ACT).LIMdomainsareconstitutedbyaconservedcysteine-andhistidine-richstructureshapedintworepeatedzincfingersfirstidentifiedintheproteinsencodedbytheLin-11,Isl-1,andMec-3genes.TheLIMdomainhasbeenshowntofunctionasaprotein-proteininteractiondomain,andhasoftenbeendescribedinassociationwithotherfunctionalproteinmotifs,suchashomeoboxandkinasedomains.FHL2seemstobeapromiscuouscoactivatorbecauseitmodulatestheactivityoftheandrogenreceptor,CRE-bindingprotein(CREB),andWT1,althoughsomedegreeofspecificityispresentbecauseitisunabletostimulateCREM-andSp1-dependenttranscription.FHL2expressionwasoriginallydescribedtoberestrictedtotheheartbutitisinducIBLeinothercelltypes.FHL2showsspecificinteractionwithbeta-catenin,whichrequirestheintactstructureofallfourLIMdomainsofFHL2andtheN-terminusplusthefirstarmADIllorepeatregionofbeta-catenin.FHL2isamuscle-specificrepressorofLEF/TCFtargetgenesandpromotesmyogenicdifferentiationbyinteractingwithbeta-catenin.MonoclonalantibodyF4B2-B11recognizestheN-terminalZn-Fingermotif,itdoesnotcrossreactinWesternBlottingwiththeFHL1andFHL3proteins.TheF4B2-B11antibodyiscrossreactivewithmouseandratFHL2.
