Description:Melanomaassociatedantigen,Human,mAbNKI/M6
Melanomaassociatedantigen(MAA)isdispersedinthecytoplasmaofmelanomacells,andismoreconcentratedinsidevacuolesandsometimesonthemelanosomes.Occasionallytheantigenisseenonthecellsurface.Theantigenisactivelyshedfromlivingcells.Althoughtheantigenisassociatedwithmelanomas,itisnotcodistributedwiththetyrosinaseactivityassociatedwithmelagonesis.TheantigenshowscodistributionwithcathepsinD,whichisaMarkerforlysosomalfunctions.ThisantibodyNKI/M6recognizesahighmolecularweightproteoglycanwithamolecularweightof;450kD(chondroitinsulfate)and250kD(coreprotein).TheantibodyNKI/M6reactswithmelanomacellsderivedfromcelllinesandshorttermculturesandreactspreferentiallywithmelanomacellsinfrozensections.NKI/M6canalsobeusedtodetectmelanomalesionsinvivo.NKI/M6showscross-reactivitywithmostnaeviandperineurium,andshowsweakreactivitywithhairfollicles.
