Themonoclonalantibody6D8recognizestheextracellulardomainofhumandesmoglein-2.Desmogleinsanddesmocollinsaremembersofthecadherinfamilyoftransmembraneproteinsthattogethermakeupthecoreofthedesmosome,astructurethatprovidestransmembranestrengthtotissuesundergoingmechanicalstress.Thedesmosomalcadherins,desmogleinsanddesmocollins,mediatecalcium-dependentcell-celladhesionbyforminghomotypicandheterotypicinteractionswithoneanother.Themultiproteindesmosomalcomplexalsoincludesthecytoplasmicdesmosomalplaqueproteinsplakoglobin,phakophilins,anddesmoplakin,whichbindtotheintracellulardomainofthedesmogleinsandfunctiontoanchorthekeratinintermediatefilamentnetworktositeofcellcellcontacts.
Inhuman,fourdesmogleinshavebeenidentified(Dsg14).Desmogleinsaresynthesizedwithasignalpeptidethatdirectsthemtotheendoplasmicreticulumandaproregionthatisremovedduringproteinprocessing.Thematureproteinincludesfourhighlyconservedextracellulardomains(EC14)andafifthmembraneproximal,morevariableECdomainthatisreferredtoasthe“extracellularanchordomain.Desmoglein-2isexpressedonvariouscellsincludingsimpleepitheliaandmyocardium,tumorsandandmanycellcultures.
Desmogleinsplaycriticalrolesincelladhesionandskinblisteringdiseases,astheyarethetargetantigensofautoimmuneantibodiesandbacterialtoxins.Desmosomaldysfunctionhasbeenimplicatedinanumberofdiseases,includingstriatepalmoplantarkeratoderma,skinfragility,andectodermaldysplasia,andmostrecentlyarrhythmicrightventricularcardiomyopathy(ARVC).
