ThemonclonalantibodyHM.2reactswiththe90kDglycoproteinIntracellularAdhesionMolecule-1(ICAM-1).TheadhesionmoleculeICAM-1belongstotheimmunoglobulinsuperfamily,C2subset;itisaligandfortheIntegrinsLFA-1andMAC-1andforCD43.ICAM-1isaessentialcomponentinmanyimmune-relatedprocesses.ICAM-1linkswithreceptorsoftheintegrinfamily,therebymediatingcell-cellinteractionsandallowingforsignaltransduction.ICAM-1interactsspecificallywithitsreceptorstoinduceareversIBLeadhesioninteraction.ForprocesseslikeTcellactivationandleucocyterecruitment,normalimmunefunctionreliesonICAM-1.Therefore,itisunderstandablethatalterationsinICAM-1structureorexpressionareassociatedwithimmunedisorders.ItisimportanttoproperlyunderstandthevariousfunctionsandregulatorymechanismsofICAM-1,theresultingdisease-relatedfailures,andthevarioustreatments.ICAM-1isatypeofintercellularadhesionmoleculecontinuouslypresentinlowconcentrationsinthemembranesofleukocytesandendothelialcells.Uponcytokinestimulation,theconcentrationsgreatlyincrease.ICAM-1canbeinducedbyinterleukin-1(IL-1)andtumornecrosisfactoralpha(TNFalpha)andisexpressedbythevascularendothelium,macrophagesandlymphocytes
