ThemonoclonalantibodyM.Ab.F11recognizesjunctionaladhesionmolecule-A(JAM-A)alsoknownasthehumanplateletF11-Receptor(F11R)orJAM-1.JAM-Aisasurfaceglycoproteinduplex(32and35kDa)belongingtotheimmunoglobulinsuperfamilyfoundonthesurfaceofhumanplateletsandatintercellularjunctionsofendothelialcellsandepithelialcells.JAM-AbelongstogetherwithJAM-C(JAM-2)andJAM-B(VE-JAMorJAM-3)toafamilyofadhesionproteinswithaV-C2immunoglobulindomainorganization.JAM-Aplaysanimportantroleintightjunctionswhereitisinvolvedincell-to-celladhesionthroughhomophilicinteractions.Itco-distributeswithothertightjunctioncomponentssuchasZO-1,7H6antigen,cingulinandoccludin.Moreover,JAM-Aplaysaroleinplateletaggregation,secretion,adhesion,spreADIng.
Intheplatelet,JAM-Aisamembraneproteininvolvedin2distinctprocessesinitiatedontheplateletsurface.Namely,,antibody-inducedplateletaggregationandsecretionbothdependentonFcgammaRIIandGPIIb/IIIaintegrin,aprocessthatmaybeinvolvedinpathophysiologicalprocessesassociatedwithcertainthrombocytopeniasandsecondly,antibodymediatedplateletadhesionindependentfromFcgammaRIIor-fibrinogenreceptorthatappearstoplayaroleinphysiologicalprocessesassociatedwithplateletadhesionandaggregation.AphysiologicalrolefortheJAM-Aproteinwasdemonstratedbyitsphosphorylationafterthestimulationofplateletsbythrombinandcollagen.ApathophysiologicalrolefortheJAM-AwasrevealedbydemonstratingthepresenceofJAM-Aantibodiesinpatientswiththrombocytopenia.AdhesionofplateletsthroughJAM-Aresultedineventscharacteristicoftheactionofcelladhesionmolecules.RecentdatasuggestsaroleforJAM-Aintheadhesionofplateletstocytokine-inflamedendothelialcellsandthusinthrombosisandatherosclerosisinducedinnon-denudedbloodvesselsbyinflammatoryprocesses.
