TheFITCconjugatedmonoclonalantibodyBV16recognizesthehumanjunctionadhesionmolecule(JAM)-A.TogetherwithJAM-C(JAM-2)andJAM-B(VE-JAMorJAM-3),JAM-AbelongstoafamilyofadhesionproteinswithaV-C2immunoglobulindomainorganizationandtheirmolecularweightisabout30-40kDa.JAMsareimportantforavarietyofcellularprocesses,includingtightjunctionassembly,leukocytetransmigration,plateletactivation,angiogenesisandvirusbinding.JAM-Aisexpressedbyendothelialandepithelialcells,platelets,neutrophils,monocytes,lymphocytesanderythrocytes.LikeallotherJAMs,JAM-Aplaysanimportantroleintightjunctionswhereitisinvolvedincell-to-celladhesionthroughhomophilicinteraction.ItcodistributeswithothertightjunctioncomponentsasZO-1,7H6antigen,cingulinandoccludin.JAM-Aalsoplaysanimportantroleinleukocytetransmigration.LeukocytetransmigrationcanbeblockedbyantibodiesandbysolubleJAM-A/Fcfusionproteins.HomophilicJAM-AinteractionsbetweenleukocytesandtheendotheliumbutalsoheterophilicinteractionsofJAM-Awiththebeta2-integrinleukocytefunction-associatedantigen-1(LFA-1)areconsideredtoactivelyguideleukocytesduringtransmigration.SeveralstudiesimplyaroleforJAM-Aintheinitiationofatherosclerosis,sinceJAM-Aisupregulatedonearlyatheroscleroticendothelium.AdhesionoftheactivatedplateletsonactivatedendotheliumismediatedbyhomophilicinteractionsofJAM-A.
