ThemonoclonalantibodyTLR3.7recognizesthe116kDahumanToll-likereceptor3(TLR3,CD283).Toll-likereceptors(TLRs)arehighlyconservedfromDrosophilatohumansandsharestructuralandfunctionalsimilarities.TLRsconstituteofafamilyofpatternrecognitionreceptors(PRRs)thatmediatecellularresponsestoalargevarietyofpathogens(viruses,bacteria,andparasites)byspecificrecognitionofso-called‘pathogen-associatedmolecularpatterns’.ActivationofTLRs,afamilyofatleast11differentmembersthatfunctioneitherashomo-orheterodimers,leadstoactivationofNFκBDependentandIFN-regulatoryfactor-dependentsignalingpathways.TLRshaveacentralroleininnateimmunityandarealsorequiredforthedevelopmentofanadaptiveimmuneresponse.TLRsareexpressedbyvariouscellsoftheimmunesystem,suchasmacrophagesanddendriticcells.TLRsareclassIreceptors,withasingleα-helixthatspansthecellmembrane.Theyrecognizeandrespondtomoleculesderivedfrombacterial,viralandfungalpathogens,suchaslipopolysaccharide(LPS)fromtheoutermembraneofGramnegativebacteria,peptidoglycanfragmentsfrombacterialcellwallsandsingle-strandedanddouble-strandedRNAfromviruses.
SomeformsofRNAandDNAfrompathogensexhibitimmutablefeaturesthatdistinguishthemfromnucleicacidsofhigherorganisms.Forexample,dsRNA,isacommonintermediateofviralreplicationandapotentindicatorofinfection.Toll-likereceptor3(TLR3)recognizesviraldouble-strandedRNAanditssyntheticanalogpolyriboinosinic:polyribocytidylicacid(poly(I:C)).TLR3isnormallylocatedinacidicendosomeswhereitsluminalectodomain(ECD)encountersdsRNAandinducestypeIinterferon(IFN),inflammatorycytokine/chemokineproductionanddendriticcell(DC)maturationviatheadaptorproteinTICAM-1(alsocalledTRIF).BasedonthedifferentsubcellularlocalizationofcytosolicRNAreceptorsandTLR3,thesereceptorsseemtoplaydistinctrolesinanti-viralimmuneresponses.

W:Totalcellularproteinwasloadedon7.5%SDS-PAGEandblottedonPDVF.Blotswereincubatedwith2μg/mlantibodyo/nat4°C(Ref.6). |
