ThemonoclonalantibodyS5/1recognizesthehumanreceptorforC5a(CD88).TheC5areceptor-isaseventransmembraneGTP-binding-protein-coupledreceptorof45kDawhichbindswithhighaffinitytohumananaphylatoxinC5a.UponC5abinding-G-protein-dependentcellular-responsesareinitiatedsuchasincreasedintracellularCa2+,granulefusionwiththecellmembrane,enzymereleaseandoxidativeburstsuggestingthatC5aRisanimportantmediatorofinflammation.TheC5areceptorisexpressedonabroadrangeofcelltypes,including-neutrophils,monocytes/macrophages,dendriticcells,astrocytesandmicroglia.
Interestingly,C5areceptorsignallinghasbeenassociatedwithseveralinflammatorydiseasesincludingrheumatoidarthritis.-
CloneS5/1wasraisedagainstasyntheticpeptidecomprisingtheN-terminaldomainoftheC5aR(Met1-Asn31).TheantibodyhasbeenshowntoinhibitthebindingofC5atoitsreceptor.
Interestingly,C5areceptorsignallinghasbeenassociatedwithseveralinflammatorydiseasesincludingrheumatoidarthritis.-
CloneS5/1wasraisedagainstasyntheticpeptidecomprisingtheN-terminaldomainoftheC5aR(Met1-Asn31).TheantibodyhasbeenshowntoinhibitthebindingofC5atoitsreceptor.
