Threepathwaysofcomplementactivationhavebeenreported:theantibody-dependentclassicalpathway,theantibody-independentalternativepathwayandthelectinpathway.Activationofeachpathwayinvolvesformationofserineproteasecomplexes,whichresultsinactivationofthecentralcomplementcomponentC3.Inthelectinpathway,mannosebinding-lectin(MBL)-associatedserineproteases(MASP)formcomplexeswithpolymericlectinmoleculeswhichareinvolvedinpatternrecognition.Uponbindingoftherecognitionmoleculestocarbohydratesonthesurfaceofmicroorganisms,MASPareconvertedtotheiractiveformsandinitiatecomplementactivation.ThreetypesofhumanMASPhavebeenreported.MASP-1,MASP-2andMASP-3.MASP-1appearstocleavethesecondcomplementcomponentC2,butnotC4.TheproteolyticactivitiesofMASP-1areinhibitedbyC1-inhibitor.FurThermoreMASP-1hasareactivityprofileverysimilartothatofthrombin.MASP-1isabletocatalysetheformationofcross-linkedfibrin.ParticipationofMASP-1incross-linkedfibrinclotformationcausesreleaseofachemotacticfactorrepresentingaBIOLOGicallysignificantactivityofMASP-1.Thealternative-splicingproductfromMASP-1geneiscalledMASP-3.MASP-1isassociatedwithsmallerMBLoligomerswhereasMASP-3isfoundonlargeroligomers.ThesubstrateofMASP-3isunknown.TheantibodyrecognizestheheavychaincommontobothMASP-1andMASP-3.
