Themonoclonalantibody5G5recognizeshumanToll-likereceptor9.Toll-likereceptors(TLRs)arehighlyconservedfromDrosophilatohumansandsharestructuralandfunctionalsimilarities.TLRsconstituteofafamilyofpatternrecognitionreceptors(PRRs)thatmediatecellularresponsestoalargevarietyofpathogens(viruses,bacteria,andparasites)byspecificrecognitionofso-called‘pathogen-associatedmolecularpatterns’.ActivationofTLRs,afamilyofatleast11differentmembersthatfunctioneitherashomo-orheterodimers,leadstoactivationofNFκB-dependentandIFNregulatoryfactor-dependentsignalingpathways.TLRshaveacentralroleininnateimmunityandarealsorequiredforthedevelopmentofanadaptiveimmuneresponse.TLRsareexpressedbyvariouscellsoftheimmunesystem,suchasmacrophagesanddendriticcells.Theyrecognizeandrespondtomoleculesderivedfrombacterial,viralandfungalpathogens.
WhereasmostTLRsareexpressedonthecellsurface,TLR9isexpressedintracellularlywithinoneormoreendosomalcompartmentsandrecognizesnucleicacids.TLR9detectsarathersubtledifferenceintheDNAofvertebratescomparedwiththatofpathogens.VertebrategenomicDNAshavemostlymethylatedCpGdinucleotideswherebacterialandviralDNAshaveunmethylatedCpGdinucleotides.TLR9undergoesrelocationfromendoplasmicreticulumtoCpG-ODN-containingendosomes.IntheseendosomesTLR9becomesafunctionalreceptorafterproteolyticcleavage.TLR9existsasapreformedhomodimerandCpG-ODNbindingpromotesitsconformationalchange,bringingthecytoplasmicTIR-likedomainsclosetoeachother.ThisallowsarecruitmentofthekeyadapterproteinMyD88whichinitiatesasignallingcascade.TheonlyhumanimmunecelltypesknowntoconstitutivelyexpressTLR9andtobeactivatedbyCpGODNarepDCsandBcells.TLR9triggeringinducesanactivationphenotypeintheBcellsandpDCs,characterizedbytheexpressionofcostimulatorymolecules,resistancetoapoptosis,andinducesTh1-typeimmuneresponseprofiles.
WhereasmostTLRsareexpressedonthecellsurface,TLR9isexpressedintracellularlywithinoneormoreendosomalcompartmentsandrecognizesnucleicacids.TLR9detectsarathersubtledifferenceintheDNAofvertebratescomparedwiththatofpathogens.VertebrategenomicDNAshavemostlymethylatedCpGdinucleotideswherebacterialandviralDNAshaveunmethylatedCpGdinucleotides.TLR9undergoesrelocationfromendoplasmicreticulumtoCpG-ODN-containingendosomes.IntheseendosomesTLR9becomesafunctionalreceptorafterproteolyticcleavage.TLR9existsasapreformedhomodimerandCpG-ODNbindingpromotesitsconformationalchange,bringingthecytoplasmicTIR-likedomainsclosetoeachother.ThisallowsarecruitmentofthekeyadapterproteinMyD88whichinitiatesasignallingcascade.TheonlyhumanimmunecelltypesknowntoconstitutivelyexpressTLR9andtobeactivatedbyCpGODNarepDCsandBcells.TLR9triggeringinducesanactivationphenotypeintheBcellsandpDCs,characterizedbytheexpressionofcostimulatorymolecules,resistancetoapoptosis,andinducesTh1-typeimmuneresponseprofiles.
