ThemonoclonalantibodyGD2.F4reactswithhumanTLR1.Toll-likereceptors(TLR)arehighlyconservedthroughoutevolutionandplayanessentialroleinrecognizingconservedmotifsfoundinvariouspathogensandinitiatinganappropriateinnateimmuneresponse.Inhuman,tenmembersoftheTLRfamilyhavebeenidentifiedastypeItransmembranesignalingreceptorscontainingmultiplecopiesofleucinerichrepeatsintheextracellulardomainandaninterleukin-1(IL-1)receptormotifinthecytoplasmicdomain.MammalianresponsivenesstomicrobialproductsmaybemediatedbycombinationsofTLRs,forexampleaco-operativeeffectisobservedbetweenTLR1andTLR2-inresponsetobacteriallipoproteins.Ontheotherhand,TLR1wasshowntohavethecapacitytoabrogateTLR4signaling.Ingeneral,TLR1isexpressedathigherlevelsascomparedtootherTLRs.ThehighestexpressionofTLR1isfoundinmonocytesbutitcanalsobeexpressedbymacrophages,dendriticcells,B,T,andNKcells.Inrecentstudies,severalhumanTLR1polymorphismshavebeenassociatedwithimpairedmycobacterialsignalingandsusceptibilitytotuberculosis.
