ThemonoclonalantibodyMHR73reactswithhumanRP105(CD180).RP105isatype1transmembraneproteinof105kDawithextracellularleucine-richrepeats(LLRs)andashortcytoplasmictail.RP105,assignedasCD180,issimilartoDrosophilaTollintheextracellularLLRs.RP105expressionislargelyrestrictedtoimmunecellsincludingCD19+Bcellsandmacrophages.HistologicalstudiesshowedthatRP105isexpressedmainlyonmatureBcellsinmantlezones.Antibody-mediated(MHR73)cross-linkingofRP105inducesresistanceagainstirrADIation-inducedapoptosis,B-cellproliferation,andup-regulationofacostimulatorymoleculeCD80,revealingRP105asapotentregulatorofB-cellactivation.InthiscontextitisimportanttonotethatRP105isassociatedwithMD1,andthatitmediatesB-cellsurfaceexpressionandLPSrecognitionandsignallingincooperationwithTLR4.MD-1seemstoberequisiteforefficientexpressionofRP105.
