Themonoclonalantibody557recognizesanepitopeofcomplementfactor5(C5)andC5a.Thecomplementsystemiscomposedofover30proteins,activatedinresponsetotissueinjury,invADIngpathogensorotherforeignsurfaces.Thecomplementpathwayscanbedividedintheactivationpathwaysandlyticpathway.
TheactivationpathwaysleadviaC3tothecleavageofthefifthcomplementcomponentC5.C5awasfirstdescribedasacleavageproductofC5withchemotacticandanaphylatoxicproperties.FurthercharacterizationrevealedthatC5aisanessentialpartoftheinnateimmuneresponseandevidencenowsuggeststhatitmayalsoplayaroleinadaptiveimmunity.ComplementfragmentC5aisa74residuepro-inflammatorypolypeptide.C5ainducessmoothmusclecontraction,increasesvascularpermeABIlity,causesdegranulationofmastcellsandbasophils,andreleaseoflysosomalenzymes.InadditionC5astimulatesthedirectedmigrationofneutrophils,eosinophils,basophilsandmonocytes.C5abindstoatleasttwoseven-transmembranedomainreceptors,C5aR(C5R1,CD88)andC5L2(gpr77),expressedubiquitouslyonawidevarietyofcellsbutparticularlyonthesurfaceofimmunecellslikemacrophages,neutrophilsandTcells.Theformerisawell-establishedreceptorthatinitiatesG-protein-coupledsignalingviamitogen-activatedproteinkinasepathways,therebybyinducingsynthesisofcytokinessuchasTNF-alpha,IL-1beta,-IL-6andIL-8.Itsinvivoblockadegreatlyreducesinflammatoryinjury.MuchlessisknownaboutC5L2,occupancyofwhichbyC5adoesnotinitiateincreasedintracellularCa(2+).ThewidespreadexpressionofC5areceptorsthroughoutthebodyallowsC5atoelicitabroadrangeofeffects.Thus,C5ahasbeenfoundtobeasignificantpathogenicdriverinanumberofimmuno-inflammatorydiseases.NowadaysC5aisalsoimplicatedinnon-immunologicalfunctionsassociatedwithdevelopmentalBIOLOGy,CNSdevelopmentandneurodegeneration,tissueregeneration,andhaematopoiesis.
Theantibody557iscapabletoinhibitthebindingofC5atotheC5areceptorthroughacompetitivemechanism,itdoesnotblockthecleavageofC5intoC5aandC5b.
