Monoclonalantibody474reactswithanepitopeonC3a.TheantibodyreactsbothwithintactC3aswithC3a.Thecomplementsystemisanimportantfactorininnateimmunity.Thethirdcomplementcomponent,C3,iscentraltotheclassical,alternativeandlectinpathwaysofcomplementactivation.ThesynthesisofC3istissue-specificandismodulatedinresponsetoavarietyofstimulatoryagents.Duringcomplementactivation,C3isproteolyticallycleavedresultinginreleaseoftheanaphylatoxicpeptideC3a.C3aisasmallpolypeptideconsistingof74aminoacids(ca.10kDa).C3aitselfisveryshort-livedandinserumcleavedrapidlyintothemorestableC3a-desArg(alsocalledacylationstimulatingprotein,ASP).C3aexertsitsfunctionthroughaspecificreceptor(C3aR),whichbelongstotheG-proteincoupledreceptorfamily.ExpressionofC3aR-hasbeenreportedinmanycelltypesincludingmyeloidandnon-myeloidcells.ExpressionofC3aRonhaematopoieticstem/Progenitorcellshasbeenshowntopromotethetrafficking/homingofthesecellstotheBM.EngagementofC3aRonDCsandTcellshasbeenshowntoup-regulatethesecellfunctions.
C3aisamediatoroflocalinflammatoryprocesses.Itinducessmoothmusclecontraction,increasesvascularpermeABIlity,andcauseshistaminereleasefrommastcellsandbasophilicleukocytes.C3aisinvolvedininflammatoryreactionsseeningram-negativebacterialsepsis,trauma,ischemicheartdisease,post-dialysissyndromeandavarietyofautoimmunediseases.AninheriteddeficiencyofC3predisposesthepersontofrequentassaultsofbacterialinfections.Inulcerativecolitis,andidiopathicchronicinflammatoryboweldisease,thedepositionofC3inthediseasedmucosahasbeenreported.
