Themonoclonalantibody755recognizesanepitopelocatedintheC-terminal360aminoacids-onthealphachainofC3,therebyrecognizingC3bandfullC3.Thecomplementsystemisanimportantfactorininnateimmunity.Thethirdcomplementcomponent,C3,iscentraltotheclassical,alternativeandlectinpathwaysofcomplementactivation.ThesynthesisofC3istissue-specificandismodulatedinresponsetoavarietyofstimulatoryagents.C3isthemostabundantproteinofthecomplementsystemwithserumproteinlevelsofabout1.3mg/ml.AninheriteddeficiencyofC3predisposesapersontofrequentbacterialinfections.C3fragmentsaredepositedintissuesatsitesofantibody-mediatedimmunopathology.Inulcerativecolitisandidiopathicchronicinflammatoryboweldisease,thedepositionofC3inthediseasedmucosahasbeenreported.Afteractivationofthecomplementsystem,certainenzymesbecomeactive,resultinginthecleavageofC3intoC3bandtheanaphylatoxinC3a.C3bbecomesattachedtoimmunecomplexesandisfurthercleavedintoiC3b,C3c,C3dgandC3f.Withinthealternativepathwayofcomplement,C3bplaysacriticalroleintheamplificationloopinitiatedbyspontaneoushydrolysisofC3.
