Description:TLR4,Human,mAbHTA125,biotinylated
Toll-likereceptors(TLR)arehighlyconservedthroughoutevolutionandhavebeenimplicatedintheinnatedefensetomanypathogens.InDrosophilatollisrequiredfortheanti-fungalresponse,whiletherelated18-wheelerisinvolvedinantibacterialdefenses.Inmammals,TLRidentifiedastypeItransmembranesignalingreceptorswithpatternrecognitioncapABIlities,havebeenimplicatedintheinnatehostdefensetopathogens.TLR4hasbeenidentifiednexttoMD-2andCD14asareceptorthatiscentraltotheinnateimmuneresponsetolipopolysaccharides(LPS)ofGram-negativebacteria.TheHTA125monoclonalantibodyreactspreferentially,especiallyinflowcytometry,withhumanTLR4(CD284)thatisassociatedwithMD-2.HTA125isaTLR4(CD284)function-blockingantibodythatisusefulforstudiesontheroleofTLR4(CD284)asareceptorforLPSinducedcytokineproductionbyTLR4bearingcells.TheantibodywasshowntoprecipitateTLR4(100kDa).TheantibodyHTA125iscrossreactivewithcanine,cynomolgusmonkey,rhesusmonkeyandmarmosetmonkey.
