Toll-likereceptors(TLR)arehighlyconservedthroughoutevolutionandhavebeenimplicatedintheinnatedefensetomanypathogens.InDrosophila,tollisrequiredfortheanti-fungalresponse,whiletherelated18-wheelerisinvolvedinantibacterialdefenses.Inmammals,TLRidentifiedastypeItransmembranesignalingreceptorswithpatternrecognitioncapABIlities,havebeenimplicatedintheinnatehostdefensetopathogens.TLR2hasbeenidentifiedasareceptorthatiscentraltotheinnateimmuneresponsetolipoproteinsofGram-negativebacteria,severalwholeGram-positivebacteria,aswellasareceptorforpeptidoglycanandlipoteichoicacidandotherbacterialcellmembraneproducts.AfunctionalinteractionbetweenTLR2andTLR6inthecellularresponsetovariousbacterialproductshasbeendiscovered.ThecurrentlyacceptedparADIgmregardsTLR2asanessentialreceptorformanyeubacterialcellwallcomponents,includinglipoproteinsandpeptidoglycan.Bacterialspeciesasdiverseasmycobacteria,spiRochetes,mycoplasma,Staphylococcusaureus,andStreptococcuspneumoniaehaveallbeenshowntomediatecellularactivationviaTLR2(CD282).
