ElafinisanepithelialproteinaseinhibitoralsoknownundervariousothernamessuchasSkin-derivedAntileukoproteinase(SKALP)andElastase-SpecificInhibitor(ESI).ElafinbelongstotheTrappingenefamilyandwasgiventhesystematicnameTrappin-2.TheTrappinfamilyisdefinedbyaN-terminaltransglutaminasesubstratedomainandaC-terminalfourdisulphidecore.Trappinshavebeensuggestedtoplayaroleintheregulationofinflammationandinprotectionagainsttissuedamageinstratifiedepithelia.Elafinisaninhibitorofleukocyteelastaseandproteinase-3andisasubstratefortransglutaminases.Theproteinisconstitutivelyexpressedinvariousepitheliaincludinghairfollicles,oesophagus,vaginaandoralcavity.Elafinisnotpresentinnormalhumanskinbutisstronglyinducedduringinflammationasinpsoriasisandwoundhealing.AntibodiestoElafincanbeusedtoevaluatetheeffectsoftreatmentofpsoriasissinceitsexpressionissignificantlycorrelatedwithclinicalscores.AntibodiestoElafinhavealsobeensuccesfullyusedtostudydifferentiationinsquamouscellcarcinomaofthehead-and-neckregion,oesophagusandskin.ItisalsoshownthatElafinpossessesantimicrobialactivityagainstgram-positiveandgram-negativebacteria.TRAB2FisanElafin/SKALPspecificantibodywhichshowsstrongreactionwithfullyprocessedElafin/SKALP(C-terminal57aa)andaweakerreactionwithhumannativefull-lengthElafin/SKALP.Theantibodycanbeusedforpurificationofhumannativefull-lengthElafin/SKALP.Fordetectionofhumannativefull-lengthElafin/SKALPweadvisetouseHM2062.
