Description:HNP1-3,Human,mAbD21
Humanneutrophildefensins(alpha-defensins)belongtothefamilyofcationictrisulfide-containingmicrobicidalpeptides.Besidesmicrobicidal,thepeptidesexertchemotactic,immunomodulatingandcytotoxicactivityandparticipateinhostdefenseandinflammation.Defensinsareimportanteffectormoleculesagainstenvelopedviruses,bacteria,fungiandprotozoa,andproteinconcentrationsrangingfrom0.5to5μMwereshowntokillawiderangeofmicrobesinvitro.DefensinshavetheABIlitytoattacksusceptIBLemicroorganismsanddestroythestructureoftargetcellmembranes.SeveralmembersofeachdefensinfamilywereshowntoactasmicrobicidesagainstdistinctGram-negativeandGram-positivebacteria,aswellasfungiandvirusesinvitro.
AzurophilicgranulesofneutrophilscontainHumanNeutrophilPeptide(HNP)1-4,whicharehighlyhomologous.Thethreeprincipalhumandefensins,HNP1-3,areuniquetoneutrophilsand-accountforabout99%ofthetotaldefensincontentofthesecells.DefensinsHNP1-3arealmostexclusivelyexpressedinneutrophils,thereforeitisconsideredaneutrophilcellMarker.Measuredamountofdefensinsis3-5microgrampermillionhumanneutrophils.WhentreatedwithHNPtheoutermembraneofEscherichiacolibecomespermeable.ThispermeabilizationfurThermorecoincidedwiththecessationofRNA,DNAandproteinsynthesis,andwithadecreasedbacterialviabilityDefensinsarerelativelyresistanttoproteolysis,lowpHandboiling.ActivationofneutrophilsleadstorapidreleaseofHNP.HNPcanbemeasuredinplasmaandotherbodyfluidsduringinfectionandinflammation.InnormalplasmaverylowlevelsofHNParepresent.Activationofpolymorphonuclearleukocytes(PMN)inplasma,asoccursduringclottingofblood,leadstoarapidreleaseofHNP.
AntiHNP1-3antibodycloneD21recognizesnaturalHNP1-3inBIOLOGicalsolutionsbymeansofELISAintissuesectionsandleukocytesmearsfixedwithethanol,methanol/acetoneorparaformaldehyde,inflowcytometryanalysisofhumanneutrophilsstainedbycellpermeabilizationmethodandinWestern-blotting(non-reduced).FurthermoretheantibodyiscrossreactivewithRhesusmonkeyandcynomolgousmacaquesHNP1-3.
