TheantibodyreactsspecificlywithHumanIL-1ra3.IL-1ra3belongstotheIL-1systemwhichincludestwoagoNISTs(IL-1alphaandIL-1beta),convertingenzymes,antagonists,tworeceptors(IL-1RIandIL-1RII)andtheIL-1receptoraccessoryprotein.ThreemolecularisoformsoftheIL-1receptorantagonist(IL-1ra)havebeenidentifiedandcloned.SecretedIL-1ra(sIL-1raorIL-1ra1)containsaclassicalleaderpeptidegivingareleasedmatureprotein.Twointracellularisoforms,icIL-1ratypeI(IL-1ra2)andicIL-1ratypeII(IL-1ra3),havenoleadersequence,thuspredictingthattheseproteinsremainintracellular.IL-1ra3mayrepresentareservoirofIL-1inhibitors,releaseduponcelldeath,whosefunctionistolimitthepro-inflammatoryactionofcelldebris.StudieshaveshownthatIL-1ra3isexpressedbyvariouscelltypesuponexposuretoinflammatorysignalsbutmostprominentlybymononuclearphagocytesandkeratinocytes.
