Themonoclonalantibody45reactswithPolymyxinB.TheantibodybindstofreePolymyxinBaswellastoPolymyxinBalreadyboundtoLPS.ThepeptideantibioticPolymyxinB(PMB)bindstobacterialendotoxin(lipopolysaccharide,LPS).TheinteractionofPMBwithLPSinvolvesionicforcesbetweenaminogroupsinPMBandnegativelychargedphosphateandcarboxylgroupsinthelipidA-Kdoregion.PMBhasrelevanceforendotoxinresearchinatleasttwoways:first,PMBreactswithLPSofmanyspeciesregardlessofvariedSEROspecificity,andthusitcanbeusedasageneralprobeformeasuringordetectingLPSorlipidA.Second,bindingofPMBtoLPSmayresultinneutralizationofthedetrimentaleffectsofLPSeitherinvitroorinvivo.Monoclonalantibody45enablesthepossibilitiestostudyquantitativelytheinteractionofPMBandLPS.
